Aruşyak Papazyan acted for the first time in 1858, at the Naum Theatre, Istanbul, under the direction of Sirabiyon Hekimyan. With her sister Ağavni Papazyan she went on tour to Izmir with the theatre group Ekşiyan Kumpanyası.

After returning to Istanbul, she played at the Orient Theatre. This theatre was opened at the Café Oriental (in the Tokatliyan Building), which had been rented for 5 years by Arakel Efendi. But despite a repertoire of comedies, melodramas and tragedies in Turkish translation, and despite it being a the best best equipped stage and auditorium, it developed such financial difficulties that it could not keep going for more than three years.

After the Orient Theatre was closed, Aruşyak Papazyan worked at the Naum Theatre again from 1863-64, this time directed by Acemyan. She acted in Turkish translated versions of plays like Medea and Francesca da Rimini, with other actors and actresses such as Mariam Dzağikyan and Dikran Sancakçıyan. Her most successful roles were Tizbe in Angelo Malpieri and Antigone.

Aruşyak Papazyan said farewell to the stage in the year 1868 after the audience had chosen Yeranuhi Karakasyan as "best-liked acress".
